Orally bioavailable nicotinic acid riboside multimers to slow the effects of aging by design
As the body ages, muscle function is impaired and muscle regeneration following injury is more challenging. This leads to muscle fatigue, impaired muscle regeneration following muscle injury, ultimately leading to a lower quality of life and mobility issues in an aging population. This impairment is, in part, due to a decrease in vascular density in muscle with age. It has been found in a previous study that there is a treatment restoring the vascular density in mouse muscle using nicotinamide mononucleotide (NMN), a metabolic precursor to NAD+. However, this compound is unstable on the shelf and in systemic circulation and requires repeated dosage to maintain high NAD+ levels. In the proposed study, Dr. Mohammad Parvez Alam will use his expertise in organic chemistry to develop NAD+ precursors with an extended release profile, intending to create the similar restorative effects as observed for NMN.
