Understanding the interplay between tryptophan and NAD metabolism during aging

Nicotinamide adenine dinucleotide (NAD) is found in all living cells and plays important roles in energy production and as a cofactor for enzymes implicated in aging. NAD levels decline with age and elevating NAD by supplementing with NAD precursors extends lifespan in both invertebrate and mammalian model systems. Interventions that target kynurenine metabolism—the pathway responsible for de novo synthesis of NAD from tryptophan—similarly extend lifespan in invertebrate models. In this project, Dr. Sutphin will examine the interplay between kynurenines and NAD in the context of aging. By understanding how kynurenine metabolism affects NAD availability, he hopes to optimally target both pathways to extend lifespan and treat age- associated disease.
