2008 AFAR Research Grant: The Anti-Aging Activity of Methylene Blue


Please give a brief summary of your AFAR research project.
Based on my previous studies, I discovered that Methylene Blue (MB) improves the quality of life in old mice and thus could delay aging. Memory functions and muscle strength in old mice fed MB were indistinguishable from young mice. These findings show that MB has promising therapeutic values for two reasons. First, the exciting behavioral and biochemical data we found in old mice. Second, MB is a drug that has been in clinical use for many decades to treat various ailments with no side effects. My AFAR research project is directed at understanding the mechanism of action of MB in order to justify clinical trials on MB to treat age-related diseases.

What problems are you addressing and what specific questions will your research seek to answer?
I want to understand the molecular and biochemical mechanisms by which MB improves cognitive function and muscle strength in old mice. At this phase, I am investigating the action of MB on the mitochondria.

What aspects of your project are most interesting from a scientific point of view?
The ability of MB to elevate the level of complex IV; a key mitochondrial enzyme. This complex is essential for energy metabolism and the prevention of the production of oxidants. Complex IV and mitochondrial dysfunction are key cytopathologies in Alzheimer's disease.

What are the implications of your research for age-related diseases and disorders?
MB has the potential to treat or delay the onset of Alzheimer's disease. MB may also delay the decline in the brain functions of Alzheimer's patients. This research will justify clinical trials specifically targeted at curing Alzheimer's disease.
